چکیده

We studied the extent to which intra-articular hyaluronic acid (HA) administered to horses with experimentally induced synovitis in the third middle carpal joint altered cartilage proteoglycans (PGs) and the biomechanical properties of the third carpal bone. Intra-articular HA (Hylartin-V) was administered to the middle carpal joints of healthy horses before and after the induction of synovitis by Escherichia coli lipopolysaccharide (LPS). Two groups of horses received intra-articular HA only, with evaluation 2 and 6 weeks later (group 2 and group 4). Two other groups (group 1 and group 3) received LPS followed by intra-articular HA and evaluation 2 and 6 weeks later. Ex vivo third carpal bone cartilage PG synthesis was measured by SO4 incorporation and third carpal bone cartilage PG content was determined by an assay for glucuronic acid. Biomechanical properties were measured by indentation testing. At the 2-week evaluation, HA did not ablate the quantitative reduction in PG synthesis and PG content in animals with experimental synovitis. However, by 6 weeks, there was a significant rebound effect in that PG synthesis and PG content were elevated in horses with experimental synovitis that had received intra-articular HA, compared with horses that had received HA only for that period of time. There were also alterations in the gel filtration profiles of newly synthesized PGs on Sepharose CL-2B among the groups. However, there were no de novo PG species associated with any of the treatments. Furthermore, we found that the biomechanical properties of third carpal bone cartilage were not altered in any of the groups. These studies showed that intra-articular HA administered to horses with experimentally induced synovitis did not provide short-term (i.e. 2-week) benefit to third carpal bone cartilage PG synthesis. However, a long-term benefit of intra-articular HA may occur after the acute effects of synovitis on PG synthesis and PG content subside.

Objectives-To use lipopolysaccharide (LPS) to create synovitis in the midcarpal joint of ponies, and to assess the morphologic, histochemical, and immunohistochemical effects of synovitis on articular cartilage of the third carpal bone. Animals-2to 3-year-old ponies, 6 control (group 1) and 6 treated (group 2). Procedure-Synovitis was induced in 1 midcarpal joint of group-2 ponies by intra-arti...

Structural characteristics of articular cartilage proteoglycan in IgG induced experimental immune synovitis.

The early changes (five weeks) in the structure of newly synthesised and endogenous articular cartilage sulphated proteoglycans were studied in lapine IgG induced experimental immune synovitis. Rabbits with immune synovitis (IS-IgG) were compared with animals with a developed hypersensitivity to IgG (I-IgG) and with non-treated normal weight matched controls. Medial and lateral femoral condyle ...

Evaluation of the inflammatory response in experimentally induced synovitis in the horse: a comparison of recombinant equine interleukin 1 beta and lipopolysaccharide.

OBJECTIVE To compare two transient models of synovitis-osteoarthritis (OA) in horses by characterizing biological changes in synovial fluid and joint tissue. METHOD Twelve skeletally mature mares were utilized in a block design. Synovitis was induced by an intra-articular injection of 100 ng recombinant equine interleukin 1 beta (reIL-1β) or 0.5 ng lipopolysaccharide (LPS) into a middle carpa...

Homing and reparative effect of intra-articular injection of autologus mesenchymal stem cells in osteoarthritic animal model

BACKGROUND This work aimed to study the homing evidence and the reparative effect of mesenchymal stem cells (MSCs) in the healing process of induced osteoarthritis in experimental animal model (donkeys). METHODS Twenty-seven donkeys were equally divided into 3 groups based on the observation period after induction of arthritis (3, 6 and 9 weeks) to achieve different degrees of osteoarthritis....

The Role of Gene Therapy in Cartilage Repair

The key principle of gene delivery to articulations by direct intra-articular injection is to release complementary DNA(cDNA)-encoding medical products that will lead to maintained, endogenous production of the gene products withinthe articulation. In fact, this has been accomplished for both in vivo and ex vivo gene delivery, using several vectors, genes, and cells in some animal models. Some c...
